Description of Orfadin

Nitisinone is the active ingredient in Orfadin, and it falls under the category of hydroxyphenyl-pyruvate dioxygenase inhibitors. You can buy Orfadin in 2mg, 5mg and 10mg strengths in oral capsule form. Each capsule contains Nitisinone along with pre-gelatinized starch. The shell of the capsule consists of titanium dioxide and gelatin, and the imprint on the capsule contains iron oxide. Do not buy Orfadin if you have an allergy to any of these ingredients, as severe reactions may take place.

Indications & Dosage of Orfadin

Orfadin is used for treating hereditary tyrosinemia type 1 (HT-1), a rare genetic condition. It is a metabolic disorder which surfaces when the patient’s body is not able to produce enough enzymes to break down proteins from food. This condition often occurs in young babies. Symptoms of the condition include fatigue, diarrhea, vomiting, nausea, sudden stomach pain, yellowing of the eyes or skin, dark urine and unusual bleeding. Orfadin works by preventing the breakdown of tyrosine amino acid in the body and preventing the buildup of toxic substances so that the liver and kidneys are safe from possible harm.

Your doctor will recommend you to take Orfadin 1 to 2mg/kg divided into two doses during the day. Initially, you will be advised to take Orfadin 1mg/kg divided into morning and evening doses. After that, the dosage can be increased depending on your response to treatment and your personal requirements.

Take Orfadin capsules at least an hour before food or 2 hours after that, as food effects are unknown. If you are giving the drug to a young child, you can open the capsule, pour its contents in some water, and give it to the child instantly.

Side Effects & Interactions of Orfadin

Fever, muscle aches, cough, throat soreness, peeling or itching of the skin, constipation, diarrhea, nausea, trouble sleeping, nervousness, thirst, and mild stomach pain can commonly occur in patients using Orfadin. However, inform your doctor at once if any of these worsen or persist. Some serious side effects associated with the drug include:

• Jaundice, identified by yellowing of the eyes or skin
• Eye pain, redness, burning or increased sensitivity to light
• Black or bloody stools
• Vision changes
• Unusual bleeding or bruising

Before you buy Orfadin, inform your doctor about all the drugs you are taking at present, especially those containing tyrosine, phenylalanine or aspartame. Other than these, do not forget to mention any herbal products or health supplements you are taking at present.

Warnings & Precautions of Orfadin

You may have to take Orfadin as part of a complete treatment plan. Your doctor will advise you to follow a diet plan too, which will restrict you from having certain foods. He/she may also advise you to take some foods in increased portion sizes.

During Orfadin treatment, your doctor will want to check your liver function. For this, he or she may decide to conduct a blood test, ultrasound or CT scan. Therefore, do not miss any scheduled appointments with your doctor and follow his or her instructions carefully.
