Description of Ogen

Ogen is a natural estrogen prepared from purified crystalline estrone. It is solubilized as the sulfate and stabilized with piperazine. It has no odor or taste and is slightly soluble in water. It is meant for oral administration. The piperazine present in Ogen ensures stability, solubility and uniform potency of estrone sulfate.

The inactive ingredients present in Ogen are lactose, hydrogenated vegetable oil wax, dibasic potassium phosphate, colloidal silicon dioxide, sodium starch glycolate, magnesium stearate, hydroxypropyl cellulose, microcrystalline cellulose and tromethamine.

Indications & Dosage of Ogen

You can buy Ogen in strengths of 0.75 mg (Ogen .625), 1.5 mg (Ogen 1.25) or 3 mg (Ogen 2.5) estropipate, where Ogen .625, Ogen 1.25 and Ogen 2.5 is the sodium estrone sulfate amount.

Ogen is indicated for:

  • Treatment of hypoestrogenism due to hypogonadism, primary ovarian failure or castration.
  • Treatment of certain symptoms of vaginal or vulval atrophy associated with menopause.
  • Treatment of vasomotor symptoms due to menopause.
  • Preventing post menopausal osteoporosis.

For the treatment of vasomotor symptoms, vulval and vaginal atrophy, the lowest dose should be administered and the medication should be discontinued as soon as possible. The following doses are to be followed :

For vasomotor syndromes :

  • One Ogen .625 mg tablet per day or two Ogen 2.5 mg tablets per day. The lowest possible dose to treat the syndrome should be chosen.

For vulval or vaginal atrophy :

  • One Ogen .625 mg tablet per day or two Ogen 2.5 mg tablets per day, depending on the tissue response of the patient. Once again, the lowest possible dose should be chosen

For the treatment of hypoestrogenism due to castration, hypogonadism, or primary ovarian failure, the following dosage is to be followed :

For female hypogonadism :

  • One Ogen 1.25 mg tablet daily to three Ogen 2.5 mg tablets daily may be given for the first three weeks of the theoretical cycle followed by a rest period of eight to ten days.

For female castration or primary ovarian failure :

  • One Ogen 1.25 mg tablet daily to three Ogen 2.5 mg tablets daily may be given for the first three weeks of the theoretical cycle followed by a rest period of eight to ten days. Depending on the severity of the symptoms and patient’s response, the dosage can be increased or reduced.

For prevention of osteoporosis :

  • One Ogen .625 tablet daily for twenty five days of a thirty one day cycle per month.

Tablets of all three dosages are standardized to provide uniform estrone activity and are scored to provide dosage flexibility. You can buy Ogen in bottles of 100 capsules. It should be stored at 25°C or below.

Side Effects & Interactions of Ogen

Ogen is known to cause various adverse effects like :

  1. Discharge from the nipples, tenderness, enlargement or pain in the breast, galactorrhea, breast cancer, fibrocystic breast changes.
  2. Increase in blood pressure, stroke, myocardial infarction, pulmonary embolism, thrombophlebitis.
  3. Abnormal bleeding patterns of the vagina, vaginitis, ovarian cancer, endometrial hyperplasia, endometrial cancer, vaginal candidiasis.
  4. Cholestatic jaundice, nausea, vomiting, pancreatitis.
  5. Erythema, pruritis, rash, hirsutism, loss of scalp hair.
  6. Asthma, changes in libido, increase or decrease in weight, headache, dizziness, chorea.

Warnings & Precautions of Ogen

Ogen can cause coronary heart disease and stroke. It has been reported to have caused endometrial cancer, breast cancer and severe hypercalcemia. Caution should be exercised while administering Ogen to a nursing mother. It is also reported to have caused withdrawal bleeding, severe nausea and vomiting. While being administered, if there are any signs of discomfort or complications, discontinue use until advised by a physician.
